Once DSM started, no you don't need to keep usb stick anymore because DSM is installed on hard drive(s)

 

in the meantime, I think it's a bad idea to remove it since starting on DSM require a Synology valid serial number which is provided by the USB stick.

 

If you reboot your PC, NAS or wathever, that will not boot on DSM.

FWIW, it IS required whenever a DSM update operation occurs and is modified by the update.

 

From the perspective of a Synology ecosystem, it is expected to be present as it cannot be removed on a real Synology box.

Yes it will run without but in the event of say a power cut it won’t reboot. Also there really isn’t a valid reason to remove it.
